Assessment of reprograming capability of human fibroblast into hepatocyte-like cell by overexpression HNF4α gene in Tet-on system

This study investigated the ability to reprogramming human fibroblast cells into hepatocyte-like by overexpressing HNF4a gene. The isolated and primary cultured from foreskin donated healthy patients. gene overexpressed through Doxycilin activatedthe vectors Tet-on-eGFP-HNF4a. after process were evaluated observing morphological changes, immunofluorescence staining assess expression of proteins specific hepatocytes such as albumin, α-fetoprotein (AFP), HNF4a, PAS evaluate glycogen store in cells. research has successfully fibroblasts. results showed that transfer, vector Tet-on-eGFP-HNF4a expressed all it did not affect growth rate cell. After two weeks, acquired morphology, they positive for AFP, staining. created with some characteristics liver fibroblasts, however, further studies are needed comprehensively them.
